Is it possible to set an effect - as intoxicated - in a NPC sheet?

For certain things if you get the wording right then FGVTT will be able to interpret those and create the correct effect when the NPC is placed onto the CT, See here for more https://fantasygroundsunity.atlassian.net/wiki/spaces/FGCP/pages/996641934/5E+NPCs+and+Encounters#NPC-Traits
You can also add effects to the NPC once they are on the CT from any of the usual sources. You can't, without extensions, add effects directly onto the NPC sheet; if that is what you are asking.

I added this trait in the NPCs sheet:
Drunk
Krudik is poisoned.
When I place the NPC on the CT, the effect is not active.

I think you may be misunderstanding a little. What happens when you add a string that FG recognises is that it creates the effect on the CT for that character. It doesn't apply the effect to anything it only creates it. So once the NPC is on the CT you'll need to apply the effect to the target by either selecting the target and then double clicking the effect or dragging the effect onto the target. So if you want your Druid to be poisoned; add the wording as you have done and then place the Druid on the CT. Then find the effect in the CT line and drag it onto the Druid

You can do this with an ext called Equipped Effects. Which name matches affects you have in the custom effect list in the top right of FG VTT.
