Can I shortcut a standard roll 4d6 and take the best 3?

Depends on the ruleset and if you are in a campaign or in the manage characters. For 5E, their is an extension that lets you do it in the campaign. FYI, it's referred to as 4d6 drop lowest (or 4d6-l).
